Objective:To investigate the efficacy of endoscopic sphincterotomy(EST)combined with lapanroscopic cholecystectomy (LC)in treating cholecystolithiasis with secondary biliary calculi. Methods:EST was performed first in 21 cases of cholecystolithiasis with secondary choledocholithiasis patients, then LC operation. Results: All 21 cases completed EST and LC operation; one of them who had made LC operation was found residual bile duct stones in cholangiography, and then was carried out EST operation. Conclusion: EST combined with LC in treating cholecystolithiasis with secondary biliary calculi is a safe and effective minimally invasive treatment mode.

Objective To evaluate the clinical effectiveness of laparoscopic cholecystectomy ( LC) combined with endoscopic sphincter-otomy ( EST) for treatment of gallstones with common bile duct stones. Methods Clinical data of 652 patients who were treated by LCs and ESTs were retrospectively analyzed. Results ESTs were successfully accomplished in 634 patients (97. 2%), and LCs were successfully ac-complished in 621 among the 634 patients (97. 9%) after ESTs. The mean length of hospitalization after LCs was 4 days (2~15 days). In all 652 patients, there were 233 cases of acute pancreatitis, including 171 cases before ESTs and 62 cases (13. 1%) after ESTs, and they were all cured after ESTs. In 621 patients after LCs, bile leakages occurred in 8 patients (1. 2%) and they were cured with intraoperative drainages. Local fluid accumulations occurred in 120 patients (19. 0%), and 118 cases of them healed without management, and the other 2 cases were cured with percutaneous tube-placement drainages
目的探讨留置鼻胆管预防十二指肠乳头括约肌切开术(EST)后胰腺炎相关因素和护理对策。方法采用回顾性研究方法,对在本院消化内科住院的60例EST术后鼻胆管引流患者进行相关因素分析,总结针对性护理措施。结果在60例EST术鼻胆管引流患者中,引流管通畅和有效引流、引流管冲洗方法、及时正确的观察与治疗、患者舒适度改变等是预防EST术后胰腺炎的有效因素。结论通过对EST术后留置鼻胆管相关因素分析,采取针对性护理措施可以控制和减少胰腺炎的发生。
Objective To discuss the relevant factors and nursing measures of indwelling nasal duct for prevention of pancreatitis after endoscopic sphincterotomy (EST).Methods Retrospective study method was adopted to analyze the relevant factors of 60 post-EST patients with endoscopic nasobiliary drainage in Department of Gastroenterology and summarize purposeful nursing measures. Results Smooth and effective drainage,flushing methods of drainage tube,Timely and correct ob-servation and treatment,and comfortable degree of the patients were relevant effective factors for prevention of post-EST pancreatitis.Conclusion Purposeful nursing measures were adopted to control and reduce the incidence of pancreatitis after analysis of relevant factors of indwelling nasal duct after EST.

Objective To Enameloplasty sealant technique versus conventional sealant technique for caries prevention. Methods 126 cases of the first permanent molars in the mandibula were sealed by EST and CST.The preservation rate of the sealant was produced after a follow-up of 3 years. Results 126 patients for more than 3 years of follow-up. Three years, the incidence of caries EST group were:0.73%, 2.38%, 3.97%;the The CST group caries incidence were:1.59%, 3.97%and 8.73%, respectively. The preservation rate of sealers EST group for three years, respectively:94.4%, 91.3%, 86.5%; preservation rate of sealers CST group three years are as follows: 91.3%, 81.7%, 65.1%; second group difference was statistically significant (P<0.05). Conclusion EST are superior in reducing the incidence of dental caries and improve the preservation rate of sealers CST, recommended should be used whenever possible in the case of conditional EST.

从NCBI的dbEST数据库中下载假臭草同族植物的EST序列,经EST序列处理及SSR位点查询,首次设计了27对假臭草EST-SSRs引物,以海南省文昌市假臭草DNA为模板,采用单因素和L16(45)正交试验对其EST-SSRs PCR反应体系进行优化。建立了假臭草EST-SSRs最佳20μLPCR反应体系:Mg2+浓度为2.75 mmol/L,模板DNA用量为35 ng,Tag DNA聚合酶为2.25 U,dNTPs浓度为0.3 mmol/L,引物浓度为0.6μmmol/L。并用14个不同来源的假臭草样本对其体系进行验证实验,结果均能获得稳定的、清晰明亮的扩增谱带。假臭草EST-SSRs标记的开发为深入研究其遗传多样性提供了基础。
In this study, we download ESTs form dbEST database and first designed 27 primer pairs for Praxelis clematidea after EST pretreatment and SSR locus identification. The sample were collected from Wenchang City in Hainan province as DNA template, and the optimization and establishment of EST-derived SSR-PCR reaction system for Praxelis clematidea was carried out by single factor test and orthogonal experiment design of L16(45). The result was indicated that the optimal EST-SSRs PCR reaction system (20μL) was contained 2.75 mmol/L Mg2+, 35 ng DNA template, 2.25 U Tag DNA polymerase, 0.3 mmol/L dNTPs and 0.6 μmol/L primer. This system was tested by 14 Praxelis clematidea from different places, which stable and clear polymorphic bands were acquired. Accordingly, the development of EST-SSRs can be used for the genetic diversity research of Praxelis clematidea.

To excavate the EST-SSR resourses of lpomoea,12812 non-redundant ESTs with the total length about 6.70 Mb were obtained by assembling 23406 ESTs from Ipomoea batatas (L.) Lam.in NCBI.With the same way,28422 unique sequences covering 17.19 Mb were generated from Ipomoea nil (L.) Roth.A total of 328 SSR loci for Ipomoea batatas with the the frequency of 2.56% and 962 SSR loci for Ipomoea nil with the frequency of 3.38% were identified by MISA.The EST-SSRs from Ipomoea batatas and Ipomoea nil had many common features.Among all the identified SSRs,dinucleotide repeats were dominant motifs and then were trinucleotide repeats.For dinucleotide repeats,AG/CT was the major motif,followed by AT/AT.And AAG/CCT was the most frequent motif among the trinucleotide repeats.The length of EST-SSRs was mainly distributed among 20-22 bp.The results indicated that these EST-SSRs with abundant repeat motif types and high potential polymorphism had high value for exploitation and utilization.

To explore the effect of herbicide 2, 4-D butyl ester on the expressions of peroxidase (POD) and esterase (EST) in Carassius auratus serum. METHODS:0.5,1.0,1.5,2.0 mL/L concentrations of 2,4-D butyl ester (according to its 24 h LC50) and 0.9%saline control group were prepared. Carassius auratus received intraperitoneal injection of 0.1 mL/g dose of each concentration. Tail blood of Carassius auratus and the supernatant of centrifugation were collected after 24 h. SDS-polyacrylamide gel electrophoresis was used to test the toxicity on POD and EST. The experimental data were analysed with ANOVA. RESULTS:POD and EST enzyme activities were inhibited at low concentrations(0.5 mL/L),induced at medium concentrations(1.0 mL/L) and decreased at high concentrations(1.5,2.0 mL/L). Compared with the control group. the effect of 2,4-d butyl ester on migration rate of EST isoenzyme was greater than POD. A large amount of Aquilaria sinensis ( Lour.) Gilg EST sequences were obtained through high -throughput sequen-cing by 454GS FLX Titanium.From these EST sequences, 956 SSRs were selected.Forty-four pairs of EST-SSR primers were designed to amplify flanking SSRs from 956 selected SSR-ESTs.Twenty-two pairs of primers were confirmed to be efficient in A. sinensis.The preliminary validation of the polymorphism of these 22 pairs of primers was further carried out in 9 samples of 3 species in Aquilaria, and the results showed that all these primers could be used in Aquilaria.This work lays a foundation for the consequent family analysis of A.sinensis and genetic diversity analysis of plants in Aquilaria.
