探讨十全大补汤对小鼠原发性肝癌的生长及血管生成的影响,阐明十全大补汤抑制原发性肝癌生长的机制。方法:构建2种荷瘤小鼠原发性肝肿瘤模型,随机分为生理盐水组、十全大补汤组。分别采用生理盐水及十全大补汤治疗10 d,摘除眼球取血,剥离瘤体,并测量其大小和质量。酶联免疫吸附测定法检测治疗后小鼠血清中血管内皮生长因子、血管抑素和内皮抑素水平。链霉菌抗生物素蛋白-过氧化物酶连结免疫组织化学法检测转移瘤微血管密度及细胞增殖。结果:小鼠肝肿瘤模型中,十全大补汤组平均肿瘤体积与生理盐水组比较明显变小(P0.01),与生理盐水组比较,十全大补汤组肿瘤组织Ki67明显降低(P0.01),MVD明显降低(P0.01)。ELISA检测显示,与生理盐水组比较,十全大补汤能够降低血清中VEGF及上调ES的表达,差异有统计学意义(P0.01)。结论:十全大补汤能明显抑制小鼠原发肝肿瘤生长,明显抑制肿瘤血管生成以及与之相关的血管生成调节因子的作用,并且可抑制肿瘤细胞增殖,促进细胞凋亡。应用十全大补汤对控制肝肿瘤的生长具有重要意义。
Objective:To explore the influence of Shi Quan Da Bu Decoction on the growth of hepatocellular carcinoma and angiogenesis,and to elucidate the mechanism of Shi Quan Da Bu Decoction inhibiting the growth of hepatocellular carcinoma.
