目的优选马钱子中士的宁与马钱子碱的提取方法与工艺。方法在单因素试验的基础上,以高效液相测定的士的宁与马钱子碱总量为指标,以乙醇浓度、乙醇用量及回流时间为影响因素,采用正交试验法优选士的宁与马钱子碱提取工艺。结果优选的最佳提取工艺为50%乙醇10倍量,回流提取3次,每次2h。结论验证实验表明该工艺合理、稳定可行。
Objective To optimize extraction process conditions of Semen Strychni. Methods With the content of Strychnine and brucine as the target,based on the single factor experiment,extraction technology of Semen Strychni was optimized by L9(34) orthogonal test,taking the concentration of ethanol,ethanol dosage and extraction time as investigated factors. Results Optimum extraction technology was:extracted 3 times with 10 times the amount of 50%ethanol,2 hours each time. Conclusion This optimized extraction technology was simple and reasonable and it was stable.

Objective To determine the content of brucine and strychnine in Zhuifeng Souguning Pills by HPLC .Methods The content of hydroxyl safflower Brucine and Strychnine was determined by HPLC ,which was conducted with a PDAD detecter and the detection wavelength set at 260nm.The column was CAPCELL PAK MG)C18(4.6mm ×250mm,5(m)with mobile phase consisted of acetonitrile-Octane sulfonate with 0.02mol/l potassium dihydrogen phosphate mixed solution of equal (with 10%phosphoric acid to adjust the ph 2.8)(23∶77)and the flow rate was 1.0ml/min.The column temperature maintained at 30℃.Results HPLC determined that hydroxyl safflower Brucine was in range from 0.0229 to 0.172μg, which presented a good linear relationship ( r=0.9991 ) .The average recovery was 96.65%, RSD was 0.94%.HPLC determined that hydroxyl safflower Strychnine was in range from 0.0423 to 0.317μg, which presented a good linear relationship ( r=0.9998) .The average recovery was 97.81%, RSD was 2.88%.Conclusion The meth

Objective:To establish the HPLC method for determination of strychnine and brucine in gubishu tablet.Methods:By the method of HPLC,Chromatographic column Agilent C18(4.6 mm × 250 mm,5μm);the mobile phase:was 250 g·L-1 methanol:750 g·L-1 buffer solution(water:acetic acid:triethylamine=230:2.4:0.3); volume lfow :1.0 mL min-1;detected wavelength:254 nm;column temperature:25℃.Results:The linear relationship of strychnine was good in 0.1208 ~ 1.4496 μg with the correlation coefficient 0.999 9, the average recovery 99.01% and RSD 0.91%.The linear relationship of brucine in 0.0948 ~ 1.1376 μg was good,with the correlation coefifcient 1, the average recovery 97.37% and RSD 1.26%.Conclusion:This method is rapid,accurate with good reproducibility,which can be used for the quality control of strychnine and brucin in gubishu tablet.

Objective:To establish new quality standard for XiaoYan ZhiTong ointment. Methods:DaiHuang (Rheum palmatum L.), HuangBo (Cortex Phellodendri Chinensis) and BanXia [Pinellia ternata (Thunb.) Breit.] were identified qualitatively by TLC, limited quantity of aconitine contained in processed CaoWu (Aconitum sp.) was de-termined, the contents of brucine and strychnine in processed MaQianZi(Strychnos nux-vomica L.) were identified by HPLC. Results:TCL spots were clear with high resolution;brucine and strychnine showed better linear relationship in the range between 0.20 and 2.0 μg and integral value of peak area (r=1.00 0), average recovery rate of brucine was 97.8%, RSD was 0.86%(n=6);average recovery rate of strychnine was 96.6%, RSD was 1.20%(n=6). Conclu-sion:The standard could be used for quality control of XiaoYan ZhiTong ointment.

Objective:To establish a method for determinate strychnine and brucine in ShiSanWei MaQianZi pill simultaneously by HPLC. Methods:HPLC was adopted under the same condition, Dikma C18(4.6 mm×250 mm, 5 μm) chromatographic column; mobile phase: acetonitrile-0.01mol/L sodium heptanesulfonate and 0.02mol/L potassium dihydrogen phosphate mix solution (regulating pH value 2.8 with 10%phosphoric acid) (10:90);detec-tion wavelength was 260nm; column temperature: 30℃; flow rate was 1.0mL/min; two kinds of ingredients in ShiSanWei MaQianZi pill were determined simultaneously. Results:Strychnine and brucine showed better linear re-lationship in the ranges between 0.262 and 1.310μg (r=0.999 8), 0.172 and 0.860μg (r=0.999 5). Average recovery rates of both were 97.96%(RSD=1.95%), and 98.06%(RSD=2.01%) respectively. Conclusion:The method, simple, exclusive and good repeatability, could be used for quality control of ShiSanWei MaQianZi pill.

Objective To establish an HPLC method for the simultaneous determination of ephedrine ,strychnine and brucine in Yaotuitong Pills .Methods The samples were separated on an Agilent C18 (250 mm × 4 .6 mm ,5μm) at 30 ℃ and detected at 210 and 254 nm .The mobile phase was consisted of acetonitrile-1 mL · L -1 phosphoric acid (12 ∶ 88 ) with a flow rate of 1 .0 mL · min-1 . Results The linear ranges of ephedrine ,strychnine and brucine were 1 .0-25 μg · mL -1 (r=0 .999 9) ,2 .0-50 μg · mL -1 (r=0 .999 8) ,2 .0-50 μg · mL -1 (r=0 .999 9) and the average recoveries were 98 .7% ,99 .1% and 97 .9% .The average contents of ephedrine ,strychnine and brucine in three batches of Yaotuitong Pills were 93 .1 ,106 .7 and 76 .8μg · g -1 .Conclusion The meth-od is sensitive ,accurate ,and can be used to control the quality of Yaotuitong Pills .

Objective To develop a RP-HPLC method for the determination of Brucine and Strychnine in Semen Strychni and its extractive of total alkaloids. Methods A chromatographic column of Licrospher C18 (4.6 mm×250 mm, 5 μm) was used with the mobile phase of acetonitrile∶0.01 mol/L sodium heptane sulfonate and 0.02 mol/L potassium dihydrogen phosphate mixed with equal amount (adjusted pH to 2.8 with 10% phosphonic acid)=27∶73, detection wavelength at 260 nm, column temperature of 30 ℃ and flow rate of 1 mL/min. Results The calibration curves of Brucine and Strychnine were both in good linearity in the ranges of 0.1-1.0 μg and 0.12-1.2 μg (r=1.000) respectively. The average recovery rates of Brucine and Strychnine were 99.88% (RSD=1.06%) and 100.06% (RSD=0.78%) respectively. Conclusion The method is realiable and accurate, which can be applied to determination of Brucine and Strychnine in Semen Strychi and its extractive.

Objective:To investigate formation and medication feature of prepared TCM medicine containing Maqianzi. Methods:14 prepared TCM medicine collected in Chinese pharmacopoeia 2010 edition were statistically analyzed. Results:78.6%of Maqianzi were powdered in 14 prepared TCM medicine, focusing on Huoxue Huayu, Tongjing Huoluo and Liqi Zhitong. Conclusion:Strychnine and brucine are the major active constituents of Maqianzi. Preparation and formation of prepared TCM medicine containing Maqianzi in Chinese pharmacopoeia 2010 edition were single;Maqianzi’s functions were unique and changeless. Clinical application of Maqianzi was confirmed due to strong toxicity. So compatibility and processing methods of Maqianzi should be studied in many ways and different levels for better effects.
