目的 评价雷公藤甲素(TPT)对诱导调节性T淋巴细胞(Treg细胞)的影响和抑制同种大鼠移植肾急性排斥反应的作用及其可能机制.方法 利用磁珠分选纯化大鼠CD4+T淋巴细胞,观察TPT抑制CD4+T淋巴细胞增殖和促进其凋亡的效应;经抗CD3单抗和抗CD28单抗共刺激与转化生长因子β1 (TGF-β1)的联合作用诱导Foxp3+ Treg细胞的分化,在分别加入不同剂量的TPT、环孢素A或撤除抗CD3单抗刺激因素的条件下,检测Foxp3表达水平的变化.建立同种大鼠肾移植急性排斥反应模型,受者腹腔注射TPT,观察移植肾组织淋巴细胞浸润和脾脏淋巴细胞亚群的变化及移植肾存活时间.结果 TPT剂量依赖性的抑制CD4+T淋巴细胞的增殖和促进活化的细胞凋亡;与环孢素A相比,TPT对TGF-β1诱导的Foxp3+ Treg细胞有增强作用;在撤除抗CD3单抗刺激刺激的条件下,TPT进一步诱导CD4+T淋巴细胞表达Foxp3; TPT减少移植肾组织CD3+、CD4+、CD25+T淋巴细胞的浸润程度,增加脾脏Foxp3+T淋巴细胞的比例,并延长同种大鼠移植肾的存活时间(P<0.05).结论 TPT可促进CD4+T淋巴细胞分化为Foxp3+ Treg细胞,可能是TPT发挥免疫抑制作用的机制之一.
Objective Triptolide (TPT),a component of the Chinese herb Triptergium wilfordii,has potent immunosuppressive and anti-inflammatory activity and is clinically used in recipients of kidney transplantation.This work aimed to investigate the effect of TPT on the differentiation of regulatory lymphocytes (Tregs) from CD4+ cells in rats and on preventing rejection in vivo.Method MACS-purified rat CD4+ cells were costimulated with anti-CD3 and anti-CD28 in the presence of TGF-beta1 to induce the expression of Forkhead box P3 (FoxP3) that was detected by ? ow cytometry.TPT and cyclosporine A (CsA) were separately added into the cultures to observe the effect on the expression of FoxP3.Kidney transplantation was performed on rats that either received no treatment or were treated with TPT after transplantation.Result TPT treatment enhanced the expression of Foxp3 in CD4+ cells,whereas CsA inhibited the Foxp3 expression.In the rat kidney transplantation model,the recipient rats treated with TPT

This article makes a study of 3-TPT paral el mechanism and uses Creo 2.0 to simulate its kinematics and set up the virtual prototype model, and then deduces the positive and inverse solving equations. By means of the functions of Matlab SimMechanics module in modeling dynamic system,3-TPT model is eatablished and the fuzzy PID control er is set up by Simulink. Combined with the control er, the dynamic simulation is carried out to obtain the velocity curve. Through the analysis and study of these curves, it comes to the conclusion that its moveability and smoothness is better. This method can be used not only to visual y analyses the movement of the paral el mechanism, but also to optimize the structure parameters,analyses the performance and give a reference to the design of fuzzy PID control er.

Taking a 3-DOF translational parallel robot mechanisms as the research object,the accurate model of parallel robot mechanisms was established by the differentiation of the kinematics. The effects of length error of driving poles,structure size change and posture change on the accuracy of robot were studied by simulations. An accuracy compensation method of 3-DOF translational par-allel robot mechanisms was proposed theoretically by using the forward error model of translational parallel mechanisms. The purpose of improving the output position and pose accuracy of parallel mechanisms was realized,with controlling telescopic link of parallel mecha-nisms. At last,the compensation method which provided a foundation for practical error and control of the mechanism was proved effec-tively by example. The new theory is meaningful and profound to optimize the mechanism design of 3TPT parallel robot.

In this paper, we introduced briefly the three major modes of reverse logistics, namely the MEMT mode where the manufacturer was responsible for the recycling, the PROT mode where the alliance contractual organization was responsible for the recycling, and the TPT mode where the TPL enterprise was responsible, then used the factor analysis model to analyze the factors influencing the selection among the modes, established the evaluation index system on the aspects of the enterprise per se, economic benefit, social benefit and technical utility as well as the reverse logistics mode evaluation and selection model based on it, and at the end applied the system in an empirical example.

As the core part of Photovoltaic ( PV) generation system ,the assessment of PV modules reliability is the great impor-tance.Starts with the structure of PV modules and analyzes the factors affecting PV modules reliability ,then discusses the charac-teristic and development of the assessment of PV modules based on historical data ,accelerated test and accelerated degradation test.Lastly,it points out that should use the assessment method based on historical data for the EVA -Si-TPT modules,and use the method based on accelerated test for the modules which are update rapidly and fail slowly .If do not get enough failure data use accelerated test ,accelerated degradation test should be considered .

The purpose of this study was to investigate the effects of creatine supplement on contractile properties of rat soleus (SOL)and extensor digitorum longus (EDL)muscle.Twenty eighteen month old male SD rats were randomly di-vided into two groups:control group (CG)and creatine supplement group (SG),with ten rats in each group.After six weeks,the content of creatine (Cr)and phosphocreatine (PCr)were deteruminated by high performance liquid chroma-tography,and contractile properties were examined in SOL and EDL muscle in virto at 30℃.The result showed that sig-nificant increases in Cr and PCr had occured in the SOL and EDL after six weeks supplementation.In SOL muscle,crea-tine supplements increased the maximum twitch tension (Pt),maximum titanic tension (Po),decreased time to maxi-mum twitch tension (TPT),but the time from maximum twitch to half relaxation decreased.In EDL muscle,creatine supplements significantly increased Pt,Po while decreased TPT.Creatine supplements increas

A method for determination of glyphosate residues in Wuyi rock tea was developed using high performance liquid chromatography tandem mass spectrometry method. The procedure is as follow,the glyphosate were extracted with water from samples, and purified using TPT. Then derivatization reaction was done in borate buffer with fluorenylmethylchloroformate(FMOC - Cl). Using 1% solution of ammonium acetate and acetonitrile as the mobile phase, mass spectrometry using electrospray ionization in positive ionization mode and multiple reaction monitoring mode. The results showed that the linear correlation coefficient r = 0. 998 when glyphosate is between1ng - 10ng/ mL. Wuyi blank samples were used in added recovery experiment at four spiked levels(n = 5),the average recovery of glyphosate were 87. 3% to 98. 0% . The hyphenated technique of high performance liquid chromatography coupled plasma mass spectrometry(HPLC-ICP-MS)was applied for the determination of four organotion compounds [trimethyltin (TMT),triphenyltin (TPT),dibutyltin (DBT),tributyltin (TBT)] in yellow rice wine. Agilent TC-C18 column was used for the separation,the mobile phase of HPLC was V(CH3CN):V(H2O):V(CH3COOH)=(70:20:10,φ), 0.05%TEA ,and pH was adjusted to 3.5. The flow rate was 0.6 mL/min.Four mix standard solution from 1 to 200 μg/L were applied for the method assessment.The experimental results indicated that, the correlation coefficient of calibration curves(R2) for each organotin compound was over 0.998 and the detection limits of the four organotin compounds were about 0.1μg/L.The Organotion Compounds were extracted by acetoni-trile(CH3CN),carbon dichloride (CH2Cl2) and methylbenzene(C7H8) from yellow rice wine respectively.And the extracted-liquid were determinated by HPLC-ICP/MS.The results showed tha

Comparing with traditional catalysts used for synthesis of polybutylene terephthalate(PBT) such as tetrabutyl titanate and tetraisopropyl titanate,effects of self-made titanium-based catalyst TY-3 on esterification time,THF content in distillate, polycondensation time and PBT properties were investigated. The results show that under same conditions,when TY-3 is used for synthesizing PBT,esterification and polycondensation time are shortest,THF content in distillate is least,chrominance of PBT is best. The best content of TY-3 is 70~80 mg∕kg. The effects of pilot test with TY-3 are better and TY-3 is expected to be applied to industrial production.

Objective To investigate the relationship between pediatric critical illness scores ( PCIS ) of severe pneumonia and blood coagulation index.Methods From August 2011 to March 2013 there were 160 cases of severe pneumonia treated in ICU of Fengrun District People’ s Hospital of Tangshan.They were recruited in experimental group.At the same period 60 healthy out-patient children undergoing physical examination in the hospital were selected in the control group.The data of two groups were analyzed.Results Compared with the control group, the experimental group had higher levels of soluble P selectin and D-two dimer but lower platelet count, and the differences were statistically significant (tsoluble P selectin =10.52, tD-two dimer =5.03, tplatelet count =11.66, all P 0.05).As the disease aggravated, the levels of soluble P selectin and D-two dimer increased, but the platelet count decreased.Group comparison showed significant difference (Fsoluble P selectin =50.45, FD-two dime
